Novel culture system via wirelessly controllable optical stimulation of the FGF signaling pathway for human and pig pluripotency

Stem cell fate is largely determined by cellular signaling networks and heavily dependent on the supplementation of exogenous recombinant proteins into culture media; however, uneven distribution inconsistent stability are closely associated with spontaneous differentiation pluripotent stem cells (PSCs) result in significant costs large-scale manufacturing. Here, we report a novel PSC system via wirelessly controllable optical activation fibroblast growth factor (FGF) pathway without need for FGF2 protein, key molecule maintaining pluripotency PSCs. Using fusion protein between cytoplasmic region FGF receptor-1 light-oxygen-voltage domain, achieved tunable, blue light-dependent human porcine Our data demonstrate that highly stimulation sufficient long-term maintenance PSCs, loss potential three germ layers. This will be cost-effective platform culture.
